MA1-41280 detects Caspase-8 in human, Rhesus monkey, and chimpanzee samples.

Target Information

Caspase 8 (CASP8) is a member of the cysteine-aspartic acid protease (caspase) family. Sequential activation of caspases plays a central role in the execution phase of cell apoptosis.
Caspases exist as inactive proenzymes composed of a pro-domain, a large protease subunit, and a small protease subunit. Activation requires proteolytic processing at conserved internal aspartic residues to generate a heterodimeric enzyme consisting of large and small subunits.

Caspase 8 is involved in programmed cell death induced by Fas and various apoptotic stimuli. Its N-terminal FADD-like death effector domain suggests interaction with Fas-interacting protein FADD. Caspase 8 has been detected in insoluble fractions of affected brain regions from Huntington disease patients, implicating a role in neurodegenerative disease.

Activated Caspase 8 triggers downstream caspases, including Caspase 9, leading to apoptosis. Overexpression induces apoptosis, which can be blocked by ICE family inhibitors. Multiple alternatively spliced transcript variants exist, though not all full-length sequences are determined.
