Product Specific Information

The eBio1A6 monoclonal antibody reacts with human thymic stromal-derived lymphopoietin receptor (TSLPR). TSLPR (~50 kDa) forms a high-affinity receptor complex with IL-7Rα (CD127) to bind TSLP. It is expressed by monocytes and CD11c⁺ dendritic cells, inducing chemokines CCL17 and CCL22. Activation of dendritic cells increases secretion of IL-4, IL-5, and IL-13, influencing CD4⁺ T cell regulation.

In mice, TSLPR deficiency alone does not affect lymphocyte counts, but combined deficiency with the common gamma-chain reduces lymphocyte numbers. The eBio1A6 antibody cross-blocks binding of another anti-human TSLPR antibody, 1D8.

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated for flow cytometric analysis of TSLPR-transfected cells at 5 µL (0.5 µg) per test.

Super Bright 780 is a tandem dye excited by the violet laser (405 nm) and emits at 780 nm. Recommended detection with a 780/60 bandpass filter. Ensure instrument compatibility.

When using multiple Super Bright-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific polymer interactions.

Compensation Notes:
Higher compensation values may occur in the violet 450/50 channel when using UltraComp eBeads ([Product # 01-2222-42]) or AbC Total Antibody Compensation Beads ([Product # A10497]). In such cases, set compensation using stained cells.

Light Sensitivity: Protect from light to prevent photo-induced oxidation.
Fixation: Samples can be stored in IC Fixation Buffer (Product # 00-8222) or 1-step Fix/Lyse Solution (Product # 00-5333) for up to 3 days at 4°C in the dark with minimal impact on signal quality.

Target Information

Thymic stromal lymphopoietin (TSLP) drives dendritic cell maturation and activation, supports regulatory T cell selection, maintains CD4⁺ T cell homeostasis, and induces allergic Th2 responses. It also promotes B cell progenitor growth and differentiation. The TSLP receptor (TSLP-R) forms a high-affinity complex with IL-7Rα, requiring STAT5 for signal transduction and inhibited by SOCS-1.
