Product Specific Information

The M5/114.15.2 monoclonal antibody reacts with mouse MHC class II (I-A and I-E) glycoproteins. It detects polymorphic determinants on B cells, monocytes, macrophages, dendritic cells, and activated T lymphocytes from specific mouse haplotypes (H-2 b, d, q, p, r, u).

• Applications Reported: Suitable for flow cytometric analysis.
• Applications Tested: Validated on stimulated mouse splenocytes at ≤0.25 µg/test (100 µL final volume).
• Recommended Use: Carefully titrate for optimal assay performance.
• Excitation: 405 nm; Emission: 780 nm; Laser: Violet.

Super Bright 780 Dye Information:

• Tandem dye excited by violet laser (405 nm), emits at 780 nm.
• Use 780/60 bandpass filter for optimal detection.
• Sensitive to photo-induced oxidation — protect from light.
• Store samples in IC Fixation Buffer or 1-step Fix/Lyse Solution up to 3 days at 4°C in dark.

Target Information

MHC class II molecules are transmembrane glycoproteins on antigen-presenting cells (macrophages, dendritic cells, B cells). They bind exogenous antigens and present them to T cells. The antigen-binding groove between alpha and beta chains is open-ended and accommodates peptides of 15–24 amino acids.

HLA and MHC antibodies are crucial in Immunopeptidomics, aiding in neoantigen identification and characterization via LC-MS/MS.
