Product Specific Information

The OX8 monoclonal antibody reacts with rat CD8 alpha, a member of the immunoglobulin superfamily that heterodimerizes with CD8 beta to form the CD8 antigen. This antigen is expressed on CD4+CD8+ thymocytes, most MHC class I-restricted peripheral T cells, and some macrophages. CD8 binds to MHC class I molecules on antigen-presenting cells and participates in T-cell receptor signal transduction via association with kinase Lck.

  Cell count: 10⁵–10⁸ cells/test (empirical optimization recommended)

Super Bright 780 is a tandem dye excited by the 405 nm violet laser and emits at 780 nm. Use a 780/60 bandpass filter. Ensure instrument compatibility.

• Compensation values may differ when using UltraComp eBeads (#01-2222-42) or AbC Total Antibody Compensation Beads (#A10497).
  In such cases, set compensation using stained cells.
• When using multiple Super Bright dye-conjugated antibodies, use Super Bright Complete Staining Buffer (#SB-4401) to reduce non-specific polymer interactions.
• Light sensitivity: Protect from light.
• Fixation: Samples can be stored up to 3 days at 4°C in the dark using IC Fixation Buffer (#00-8222-49) or 1-step Fix/Lyse Solution (#00-5333-57).

Target Information

Cluster of differentiation 8 (CD8) is a type I transmembrane glycoprotein in the immunoglobulin receptor family. It plays a critical role in signal transduction, T-cell differentiation, and activation. CD8 functions as a co-receptor with the T-cell receptor (TCR) for MHC class I molecules and is expressed on T cells, NK cells, monocytes, and dendritic cells. CD8α binds MHC-I, triggering Lck kinase recruitment and cytotoxic T lymphocyte (CTL) activation for pathogen and tumor clearance. Differentiation of naive CD8+ T cells into CTLs is enhanced by IL-2, IL-12, and TGF-β1.
