Product Specific Information

The eBioCB16 monoclonal antibody recognizes CD16 (Fc gammaRIII), a low-affinity receptor for IgG (50–80 kDa).
CD16 includes two genes: CD16A (FcγRIIIA) in macrophages and NK cells, and CD16B (FcγRIIIB) in neutrophils.
Two polymorphisms (NA-1, NA-2) exist, with NA-2 homozygotes showing lower phagocytic capacity.
CD16 binds IgG immune complexes, preferentially IgG1 and IgG3, initiating signal cascades for ADCC, phagocytosis, degranulation, and proliferation.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-diluted and tested with human peripheral blood cells at 5 µL (0.6 µg)/test (100 µL final volume).

Cell number range: 10⁵–10⁸ cells/test.

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., PI, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains with NovaFluor dyes.
• Use CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) for optimal staining.
  • Add 5 µL per sample to antibody cocktails/master mixes before combining with cells.
  • For single-color controls: 5 µL per 100 µL of cell sample (10³–10⁸ cells).

NovaFluor conjugates utilize Phiton™ technology for engineered fluorescent signatures minimizing spillover and spread.

Target Information

CD16 (FCGR3A) encodes a receptor for the Fc portion of IgG, involved in immune complex clearance and antibody-dependent cytotoxicity.
FCGR3A is expressed on NK cells (transmembrane form), while FCGR3B is expressed on neutrophils (PI-anchored form).
Mutations may cause Immunodeficiency 20 and increase susceptibility to viral infections and autoimmune diseases (e.g., lupus).
Alternative splicing produces multiple isoforms.
