Product Specific Information

The eBioCB16 monoclonal antibody recognizes CD16 (Fc gammaRIII), a low-affinity receptor for IgG (50–80 kDa).
CD16 includes two similar genes:

  • CD16A (FcγRIIIA): hetero-oligomeric, expressed in macrophages and NK cells
  • CD16B (FcγRIIIB): monomeric, GPI-anchored, expressed in neutrophils

Polymorphisms NA-1 and NA-2 affect phagocytic capacity. CD16 binds IgG immune complexes, preferentially IgG1 and IgG3, and triggers ADCC, phagocytosis, degranulation, and proliferation.

Each product includes:

  • 1 vial of NovaFluor conjugate
  • 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-diluted and tested on normal human peripheral blood cells at 5 µL (0.6 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).

Important Notes:

  • NovaFluor dyes are not compatible with DNA intercalating viability dyes (PI, 7-AAD, DAPI).
  • Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assays.
  • CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining.
    • Add 5 µL per sample or per 100 µL of cell sample for single-color controls.
    • Recommended for all NovaFluor antibody panels.

NovaFluor conjugates utilize Phiton™ technology, providing engineered fluorescent signatures optimized for minimal spillover and spread.

Target Information

CD16 (FcγRIII) encodes a receptor that recognizes the Fc portion of IgG, involved in immune complex clearance and antibody-dependent cytotoxicity.

  • FCGR3A: expressed on NK cells, transmembrane form
  • FCGR3B: expressed on neutrophils, PI-anchored form

Mutations are linked to:

  • Immunodeficiency 20
  • Recurrent viral infections
  • Systemic lupus erythematosus
  • Alloimmune neonatal neutropenia

Alternative splicing produces multiple isoforms.
