Product Specific Information

The SK7 monoclonal antibody reacts with human and chimpanzee CD3e, a 20 kDa subunit of the TCR complex. The epsilon chain is essential for proper assembly, trafficking, and surface expression of the TCR complex. CD3 is expressed by thymocytes and mature T cells. SK7 and UCHT1 antibodies recognize overlapping epitopes.

Each product includes:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-diluted and tested by flow cytometry of normal human peripheral blood cells. Use 5 µL (0.8 µg) per test (100 µL final volume). Cell count: 10⁵–10⁸ cells/test.

Note:
NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., PI, 7-AAD, DAPI). Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.

CellBlox Plus Blocking Buffer (Cat. No. C001T06F01):
Required for optimal staining with NovaFluor conjugates. Add 5 µL per sample or per 100 µL of cell sample (10³–10⁸ cells).

Target Information

The CD3 complex transduces antigen-recognition signals into T cells and regulates TCR surface expression. CD3 subunits (gamma, delta, epsilon, zeta) are structurally related immunoglobulin superfamily members encoded on chromosome 11. CD3 is expressed on 68–82% of peripheral blood lymphocytes and 65–85% of thymocytes, absent on B and NK cells. Defects in CD3 genes are linked to CD3 immunodeficiency.
