Description:
The SK7 monoclonal antibody reacts with human and chimpanzee CD3e, a 20 kDa subunit of the TCR complex. Along with other CD3 subunits (gamma and delta), the epsilon chain is required for proper assembly, trafficking, and surface expression of the TCR complex. CD3 is expressed by thymocytes in a developmentally regulated manner and by all mature T cells. The SK7 and UCHT1 monoclonal antibodies cross-block binding, suggesting recognition of overlapping epitopes.
Each product contains:
- 1 vial of NovaFluor conjugate
- 1 vial of CellBlox Plus Blocking Buffer
Applications Reported: Flow cytometric analysis.
Applications Tested: Pre-diluted and tested by flow cytometric analysis of normal human peripheral blood cells. Use 5 µL (0.8 µg) per test (100 µL final volume). Cell number: 10⁵–10⁸ cells/test.
Note:
NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI). Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.
CellBlox Plus Blocking Buffer (Cat. No. C001T06F01):
Contains formulation improvements over the original CellBlox. Required for optimal staining with NovaFluor conjugates.
- Add 5 µL per sample to antibody cocktails before combining with cells.
- For single-color controls: 5 µL per 100 µL cell sample (10³–10⁸ cells).

Target Information
The CD3 subunit complex is crucial for transducing antigen-recognition signals into the cytoplasm of T cells and regulating TCR surface expression. It includes subunits CD3 gamma, delta, epsilon, and zeta, all encoded on human chromosome 11. These subunits associate with cytoplasmic signaling molecules via double tyrosine-based motifs. CD3 is expressed on 68–82% of normal peripheral blood lymphocytes, 65–85% of thymocytes, and Purkinje cells in the cerebellum, but not on B or NK cells. Decreased T lymphocyte percentages may be observed in autoimmune diseases. Mutations in CD3 genes can cause CD3 immunodeficiency.
