Product Specific Information

This SRBCZX monoclonal antibody recognizes STAT5 when phosphorylated on tyrosine 694. STAT proteins are activated by cytokine receptor-associated JAK kinases, leading to nuclear translocation and transcriptional regulation. Phosphorylation at Tyr694 is essential for dimer formation and DNA binding.

  • Specificity Determination: ELISA and flow cytometry
  • Applications Reported: Intracellular staining followed by flow cytometric analysis
  • Applications Tested: Pre-titrated and validated for intracellular staining of stimulated human peripheral blood cells (5 µL/test, 100 µL final volume, 10⁵–10⁸ cells/test)
  • Recommended Protocol: Protocol C (Two-step: Fixation/Methanol)
    • Protocol A and B not suitable for this antibody

Light Sensitivity: Protect vial and stained samples from light.
Fixation: Samples can be stored up to 3 days in the dark at 4°C using IC Fixation Buffer (Product #00-8222) or 1-step Fix/Lyse Solution (Product #00-5333).
Excitation/Emission: 488–561 nm / 775 nm; compatible lasers: Blue, Green, Yellow-Green.
Filtration: 0.2 µm post-manufacturing filtered.


Target Information

STAT5 alpha is a transcription factor activated by cytokines and growth factors. Upon phosphorylation, it forms dimers and translocates to the nucleus to regulate gene expression. It mediates responses to IL2, IL3, IL7, GM-CSF, erythropoietin, and growth hormones. STAT5 alpha activation is implicated in tumorigenesis and anti-apoptotic signaling. STAT5 alpha and STAT5 beta share 96% sequence similarity and are co-expressed in most tissues.
