Description:
The eBio1D3 monoclonal antibody reacts with mouse CD19, a 95 kDa transmembrane glycoprotein expressed by B cells during all stages of development, except plasma cells. Follicular dendritic cells also express CD19. CD19 forms a complex with CD21, CD81, and MHC class II, associating with the B cell receptor (BCR) to mediate signaling that induces tyrosine phosphorylation, calcium flux, and proliferation.

Applications Reported:
This antibody is reported for use in flow cytometric analysis.

Applications Tested:
Tested by flow cytometric analysis of mouse splenocytes. Recommended ≤0.5 µg/test (100 µL final volume). Optimal titration required for specific assay conditions.

Fluorophore Information:
Super Bright 645 is a tandem dye excited by the violet laser (405 nm) and emits at 645 nm. Recommended detection filter: 660/20 nm bandpass. Ensure instrument compatibility.

Staining Recommendations:
When using multiple Super Bright dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific interactions.

Light Sensitivity:
Protect vial and stained samples from light to prevent photo-induced oxidation.

Fixation:
Samples can be stored up to 3 days at 4°C in the dark using IC Fixation Buffer (Product # 00-8222) or 1-step Fix/Lyse Solution (Product # 00-5333) with minimal effect on brightness and FRET efficiency.

Target Information

CD19 is a member of the immunoglobulin superfamily with two Ig-like domains. It is expressed on 100% of peripheral B cells and is also found on some myeloid leukemia cells. CD19 serves as a key regulator of B cell proliferation and differentiation, lowering the threshold for antigen receptor stimulation. Mutations in CD19 can cause hypogammaglobulinemia, while overexpression leads to B cell hyperactivity.
