Description:
This CD25-4E3 monoclonal antibody reacts to human CD25 (IL-2 receptor alpha). CD25 is expressed on activated T cells (including regulatory T cells), B cells, and macrophages. It forms a high-affinity IL-2 receptor complex with CD122 and CD132, mediating IL-2 signaling involved in lymphocyte proliferation and survival. CD25 serves as a marker for Hodgkin’s disease and is associated with autoimmune conditions such as Type I diabetes.

This antibody is cross-reactive to rhesus monkey.

Applications Reported: Flow cytometric analysis.
Applications Tested: Pre-diluted and validated for flow cytometry using normal human peripheral blood cells. Recommended usage: 5 µL (0.25 µg) per 10⁵–10⁸ cells/test in 100 µL volume.

Fluorochrome Information:
Super Bright 645 is a tandem dye excited by the 405 nm violet laser and emits at 645 nm. Recommended detection filter: 660/20 bandpass.

Staining Buffer Recommendation:
When using multiple Super Bright-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific polymer interactions.

Light Sensitivity:
Sensitive to photo-induced oxidation; protect from light.

Fixation:
Samples can be stored up to 3 days at 4°C in the dark using:

• IC Fixation Buffer (Product # 00-8222-49)
• 1-step Fix/Lyse Solution (Product # 00-5333-57)

CD25 (IL2RA)
CD25 encodes the IL-2 receptor alpha chain, part of a heterotrimeric receptor complex (with IL2RB and IL2RG) that mediates IL-2 signaling. This cytokine receptor regulates T and B lymphocyte proliferation, differentiation, and apoptosis. IL-2 promotes B-cell proliferation, enhances NK cell activity, and inhibits granulocyte-macrophage colony formation. Mutations are associated with IL-2 receptor alpha deficiency and immune dysregulation.
