Target Information
p53은 종양 억제 단백질로, 세포 스트레스에 의해 활성화되는 서열 특이적 전사 인자입니다. DNA 손상이나 뉴클레오타이드 결핍 시 세포 주기 정지 또는 세포 사멸을 유도합니다. p53은 스트레스 신호에 반응하여 p21waf1 등의 유전자를 활성화하며, G1 또는 G2/M 단계에서 세포 주기를 조절합니다.
p53 구조는 N-말단 전사활성 도메인, 중앙 DNA 결합 도메인, 올리고머화 도메인, C-말단 조절 도메인으로 구성됩니다. Ser15 및 Ser392의 인산화는 p53의 활성화 및 종양 형성과 연관됩니다.
p53은 정상 세포에서는 낮은 수준으로 존재하지만, 다양한 변형된 세포주에서는 높은 수준으로 발현되어 암 발생에 기여할 수 있습니다. TP53 유전자의 변이는 여러 인간 암에서 발견되며, Li-Fraumeni 증후군과 같은 유전적 암 소인 가족에서도 보고되었습니다.
