Target Information
KRAB (Kruppel-associated box) 도메인은 약 45 아미노산 길이의 전사 억제 도메인으로, 다수의 전사 인자에서 발견됩니다. 인간 유전체에는 220개 이상의 KRAB-zinc finger protein (KRAB-ZFP) 유전자가 존재하며, 이들은 KAP-1 (KRAB-associated protein 1)과 상호작용하여 전사 억제를 매개합니다.
KAP-1은 835 아미노산로 구성된 단백질로, RING finger, B boxes, PHD finger를 포함합니다. KAP-1은 KRAB 도메인 전사 인자 및 HP1(heterochromatin protein 1)과 상호작용하며, KRAZ1을 centromeric heterochromatin으로 유도하여 전사 억제를 조절합니다. HP1 결합 능력을 잃은 KAP-1 변이는 KRAZ1의 무작위 분포 및 강한 전사 활성화를 유도합니다.
