Target Information
The differentiation and activation of monocytes (MO) and monocytic cells is modulated by 1α,25-dihydroxyvitamin D3 (Vitamin D3). Using mRNA Differential Display technology, a gene induced in human blood monocytes cultured with Vitamin D3 was identified. This gene encodes a 16.5 kD protein homologous to yeast MMS2p and human CROC-1, defining a conserved Ubc-like protein family. The protein, Ubiquitin E2 variant (UBE2V2 or MMS2), works with Ubc13 in RAD6/RAD18-dependent postreplicative DNA repair and K63-linked polyubiquitin chain assembly. These UEV/Ubc complexes are involved in DNA repair and IκBα kinase activation. Crystal structure analysis confirms that the Mms2/Ubc13 heterodimer is essential for DNA repair.
