Target Information
SPINK1 is a trypsin inhibitor whose physiological function is to prevent premature activation of zymogens within the pancreas.
Defects in SPINK1 are associated with pancreatitis (PCTT, MIM:167800), characterized by calculi in pancreatic ducts and severe abdominal pain.
Mutations also cause susceptibility to tropical calcific pancreatitis (TCP, MIM:608189), an idiopathic, juvenile, nonalcoholic chronic pancreatitis prevalent in tropical regions.
TCP can be linked to fibrocalculous pancreatic diabetes (FCPD) and differs from alcoholic pancreatitis by earlier onset, pancreatic calcification, insulin-dependent but ketosis-resistant diabetes, and high incidence of pancreatic cancer.
