Product Specific Information

The 2H7 monoclonal antibody reacts with human CD20, a 33–36 kDa transmembrane protein expressed on developing and mature B cells but not plasma cells. Low-level expression may occur on a subset of mature T cells. CD20 plays a role in B-cell activation.

• Applications Reported: Flow cytometric analysis
• Applications Tested: Pre-diluted and validated for flow cytometric analysis of normal human peripheral blood cells. Recommended at 5 µL (0.5 µg) per test (100 µL final volume).
• Excitation / Emission: 405 nm / 702 nm (Violet Laser)
• Recommended Filter: 710/50 bandpass

Super Bright 702 is a tandem dye excited by the violet laser and emits at 702 nm. Use Super Bright Complete Staining Buffer (Product # SB-4401) when combining multiple Super Bright conjugates to minimize non-specific polymer interactions.

Light Sensitivity: Protect vial and stained samples from light.
Fixation: Samples can be stored up to 3 days at 4°C in the dark using IC Fixation Buffer (Product # 00-8222) or 1-step Fix/Lyse Solution (Product # 00-5333) with minimal impact on brightness and FRET efficiency.

Target Information

CD20 is a non-glycosylated surface phosphoprotein (33–37 kDa) expressed on mature and malignant B cells, some T lymphocytes, and follicular dendritic cells. It regulates calcium conductance, cell cycle progression, and B-cell proliferation. CD20 remains on the cell surface after antibody binding, making it a valuable target for therapeutic B-cell depletion.
Diseases associated with CD20 dysfunction include Ms4a1-related common variable immune deficiency.
