Product Specific Information

Description:
The ICRF44 monoclonal antibody reacts with human CD11b, a 165 kDa adhesion molecule expressed on monocytes, granulocytes, activated lymphocytes, and a subset of NK cells. CD11b forms a heterodimer with CD18 and functions as a receptor for ICAM family members and iC3b, mediating crucial cell-cell and cell-matrix interactions.

Applications Tested:

• Pre-diluted and tested on normal human peripheral blood cells
• Recommended use: 5 µL (0.125 µg) per test in 100 µL final volume
• Suitable for 10^5–10^8 cells/test

Fluorophore Information:
Super Bright 702 is a tandem dye excited at 405 nm and emits at 702 nm. Recommended detection using a 710/50 bandpass filter. Protect from light due to photo-induced oxidation sensitivity.

Staining Buffer Recommendation:
When using multiple Super Bright-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific polymer interactions.

Fixation Compatibility:
Samples can be fixed using IC Fixation Buffer (Product # 00-8222) or 1-step Fix/Lyse Solution (Product # 00-5333) and stored up to 3 days at 4°C in the dark with minimal impact on brightness.

Target Information

CD11b (integrin alpha-M, ITGAM) is a 165 kDa adhesion molecule that forms a heterodimer with CD18 (integrin beta-2). The CD11b/CD18 complex, also known as Mac-1 or CR3, acts as a receptor for ICAMs, iC3b, fibrinogen, and other ligands, facilitating immune cell adhesion and migration.
CD11b is expressed on monocytes/macrophages, granulocytes, activated lymphocytes, NK cells, dendritic cells, and microglia. It is used as a marker for microglia and macrophage maturation.
Dysfunction of CD11b is associated with autoimmune conditions such as systemic lupus erythematosus.
