Product Specific Information

The eBioCB16 monoclonal antibody recognizes CD16 (Fc gammaRIII), a low-affinity receptor for IgG (50–80 kDa).
CD16 includes two gene variants:

• CD16A (FcγRIIIA): membrane-anchored form in macrophages and NK cells
• CD16B (FcγRIIIB): GPI-anchored form in neutrophils

Two polymorphisms (NA-1, NA-2) exist; NA-2 homozygotes show reduced phagocytic capacity.
CD16 binds immune complexed IgG, preferentially IgG1/IgG3, and initiates signaling cascades leading to ADCC, phagocytosis, degranulation, and proliferation.

Applications

• Reported for flow cytometric analysis
• Pre-diluted and tested for normal human peripheral blood cells
• Recommended usage: 5 µL (0.06 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test)

Target Information

CD16 (FCGR3A) encodes a receptor recognizing the Fc portion of IgG, involved in:

• Clearance of immune complexes
• Antibody-dependent cellular cytotoxicity (ADCC)
• Viral infection enhancement

Expression:

• FCGR3A: NK cells, membrane-anchored via transmembrane peptide
• FCGR3B: Neutrophils, anchored via phosphatidylinositol linkage

Clinical relevance:
Mutations linked to Immunodeficiency 20, Herpes Zoster susceptibility, Systemic lupus erythematosus, and alloimmune neonatal neutropenia.
Alternative splicing produces multiple isoforms.
