Description:
The PC10 antibody reacts with human PCNA, a protein involved in the control of eukaryotic DNA replication and localized in the nucleus.

Applications Reported:
Reported for use in flow cytometry, immunocytochemistry, western blotting, and immunohistochemistry of formalin-fixed paraffin-embedded tissues.

Applications Tested:
Tested by IHC (paraffin) using high pH antigen retrieval at 20 µg/mL. Optimal titration is recommended for specific assay conditions.

Handling Instructions:
Before use, briefly centrifuge the conjugate solution and use only the supernatant to remove potential aggregates, minimizing nonspecific background staining.


Target Information

PCNA (Proliferating Cell Nuclear Antigen):
A key factor in DNA replication, excision repair, and mismatch repair pathways. PCNA interacts with p21, Fen1, XPG, and DNA cytosine 5-methyltransferase (MCMT). It serves as a marker for proliferating cells, particularly in early G1 and S phases, and is widely used in tumor proliferation studies. PCNA functions as a homotrimeric cofactor of DNA polymerase delta, enhancing DNA synthesis processivity. Upon DNA damage, PCNA becomes ubiquitinated and participates in the RAD6-dependent repair pathway. Two transcript variants encode the same protein, and pseudogenes exist on chromosomes 4 and X.
