Product Specific Information

The KP1 antibody reacts with human CD68, a member of the sialomucin family related to lysosomal-associated membrane proteins (LAMPs) and scavenger receptors. CD68 is primarily an intracellular protein localized in late endosomes, with smaller amounts on the surface of myeloid-derived cells such as monocytes, macrophages, dendritic cells, neutrophils, basophils, mast cells, myeloid progenitors, and a subset of CD34+ bone marrow progenitor cells.

Although its exact function is not fully elucidated, CD68 is believed to play roles in antigen processing/presentation and in lysosomal membrane protection. Expression has also been reported in activated T cells, subsets of B lymphocytes, and fibroblasts (possibly due to conserved epitopes).

Applications Reported: Flow cytometry, immunoprecipitation, western blotting, and immunohistochemistry (frozen and paraffin-embedded tissues).
Applications Tested: Validated for IHC (paraffin) using high pH antigen retrieval at 8 µg/mL. Optimal dilution should be determined experimentally.

Note: Before use, briefly centrifuge the conjugate solution and use only the supernatant to minimize background due to aggregates.

Target Information

CD68 (Macrosialin) is a 110 kDa integral membrane glycoprotein primarily expressed in lysosomes of monocytes and macrophages, and to a lesser extent in dendritic cells and granulocytes. It may be involved in phagocytosis and lysosomal metabolism as well as cell-cell and cell-pathogen interactions.

CD68 expression is observed in interdigitating reticulum cells, histiocytic lymphomas, acute myeloid leukemia (AML), and granulocytic sarcomas. Elevated CD68 levels have been found in CD34+ cells in several malignancies, including AML.
