이 단백질은 여포 수지상세포(FDC)와 성숙 B세포, 일부 상피세포에서 발현되며, 정상 림프절 및 편도 조직의 FDC matrix 식별과 비정상 조직의 판별에 유용합니다.

CD21 (Complement receptor 2, CR2, C3D receptor, EBV receptor)은 C3 보체 단편과 결합하며, 면역복합체의 섭취와 유지, 기억 B세포의 생존, T-의존 항원에 대한 체액성 면역 반응 유지에 중요한 역할을 합니다.
또한 Epstein-Barr virus의 주요 수용체로 작용하며, 프리온의 여포 수지상세포 표적화 및 신경 침투를 촉진합니다.
가용형 CD21(sCD21)은 림프구 표면에서 절단되어 리간드 결합 능력을 유지합니다.
