Target Information
TID1은 Drosophila의 tumor suppressor lethal (2) tumorous imaginal discs, l(2)tid의 인간 상동체로, 인간 Tid-1의 두 가지 미토콘드리아 매트릭스 국소화 스플라이스 변형체인 hTid-1S와 hTid-1L을 인코딩합니다. 이들은 DnaJ 단백질 계열의 보존된 구성원으로, 미토콘드리아 Hsp70의 코샤페론으로 작용합니다.
이 단백질들은 Hsp70 샤페론과 결합하여 ATPase 활성을 활성화시키는 J 도메인을 포함합니다. hTid-1L의 발현은 DNA 손상 유발제(예: mitomycin-C, TNF-α)에 의해 유도되는 apoptosis를 증가시키며, J-domain 변이체는 apoptosis를 억제할 수 있습니다. 반면, hTid-1S의 J-domain 변이체는 apoptosis를 증가시킵니다.
두 스플라이스 변형체의 발현은 미토콘드리아에서 cytochrome c 방출 및 caspase 3 활성화에 영향을 미치며, caspase 8 활성화에는 영향을 주지 않습니다. 따라서 이 두 단백질 변형체는 각각 항- 및 친-apoptotic 효과를 발휘하며, 그 상대적 풍부도는 미토콘드리아의 apoptosis 신호 조절에 영향을 줄 수 있습니다.
