Product Specific Information

The eBio7D4 antibody reacts with mouse CD25, the 55 kDa interleukin-2 receptor α chain (IL-2Ra). CD25 is expressed by early progenitors of the T and B lineage, as well as by activated mature T and B lymphocytes.
CD25 binds IL-2 with low affinity alone, but when associated with CD122 (IL-2Rβ) and CD132 (common γ chain), forms the high-affinity IL-2 receptor. CD25 is involved in lymphocyte differentiation and activation/proliferation.

The epitopes recognized by eBio3C7, PC61.5, and eBio7D4 are distinct, allowing sequential depletion and detection. eBio7D4 may block IL-2 binding, which can be overcome by additional IL-2.
For in vivo studies, the monoclonal antibody PC61.5 (Product #16-0251) is recommended.

Applications Tested: Tested on mouse splenocytes at ≤0.125 µg/test.
Cell number range: 10⁵–10⁸ cells/test. Antibody titration recommended for optimal performance.

Target Information

CD25 (IL2 receptor alpha chain/IL2RA) is a cytokine receptor involved in T and B lymphocyte proliferation.
The receptor complex (IL2RA, IL2RB, IL2RG) forms the high-affinity IL-2 receptor.
Homodimeric IL2RA chains yield low-affinity receptors, while IL2RB homodimers yield medium affinity.
IL2 expression in mature thymocytes is monoallelic, regulating gene expression precisely. IL2 is mainly produced by mature T cells and functions as a growth, differentiation, and cell death regulator.
IL-2 enhances B cell proliferation, NK cell activity, and inhibits granulocyte macrophage colony formation.
Mutations in IL2RA are associated with interleukin 2 receptor alpha deficiency and immune dysfunction.
