Target Information
Protein p21 is a tumor suppressor protein that inhibits Cyclin-dependent kinases (CDK) and is transcriptionally activated by p53.
It plays a crucial role in the cellular response to genotoxic stress and is a major transcriptional target of p53.
In the nucleus, p21 binds and inhibits CDK1 and CDK2, blocking the transition from G1 to S phase or G2 to mitosis after DNA damage, enabling DNA repair.
In the cytoplasm, p21 has anti-apoptotic effects by inhibiting caspase 3 and apoptotic kinases ASK1 and JNK.
Thus, p21 exhibits dual functions in carcinogenesis — acting both as a tumor suppressor and as an oncogene under certain conditions.
