The monoclonal antibody CF-145 recognizes human S100A8, also known as MRP-8 or Calgranulin-A. S100A8 is a calcium-binding protein in the S100 family, expressed by neutrophils, monocytes, macrophages, granulocytes, and epithelial cells during dermatoses such as psoriasis.
It forms a heterodimer with S100A9 (calprotectin) and has antimicrobial and pro-apoptotic properties, acting as a pro-inflammatory mediator in acute and chronic inflammation. Elevated S100A8 levels are observed in inflammatory disorders including cystic fibrosis, rheumatoid arthritis, and chronic bronchitis, as well as in various tumors (skin, breast, colon, pancreatic, ovarian).

Applications Tested:
This CF-145 antibody has been pre-titrated and tested by intracellular staining of normal human peripheral blood cells using the Intracellular Fixation & Permeabilization Buffer Set (Product # 88-8824-00) and the BestProtocols® “Protocol A: Two step protocol for cytoplasmic intracellular proteins.”
Use at 5 µL (0.06 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).

Target Information

The encoded protein is part of the S100 family, containing two EF-hand calcium-binding motifs. S100 proteins are localized in the cytoplasm and/or nucleus and regulate processes such as cell cycle progression and differentiation.
S100 genes (at least 13 members) are clustered on chromosome 1q21. This protein may inhibit casein kinase and act as a cytokine. Altered expression is associated with cystic fibrosis.
