Target Information
KLOTHO is a systemic anti-aging hormone within the glycosidase1 superfamily. It encodes a type I membrane protein abundant in the kidney and brain. In mice, deficiency in KLOTHO expression results in phenotypes resembling human aging such as arteriosclerosis, osteoporosis, skin atrophy, growth retardation, short life span, and infertility. Overexpression of KLOTHO extends life span by inhibiting insulin/IGF1 signaling. KLOTHO regulates calcium/phosphorus homeostasis by inhibiting active vitamin D synthesis and acts as a potential tumor suppressor.
