Target Information
BCL-2 is a key regulator of apoptosis that functions to either inhibit or promote cell death. Members of the BCL-2 family are characterized by their ability to dimerize, modulating apoptotic activity. For example, Bag1 forms a heterodimer with BCL-2, enhancing its anti-apoptotic effect. Conversely, Bax and Bak promote cytochrome c release from mitochondria, initiating apoptosis. Constitutive expression of BCL2, such as via translocation to the Ig heavy chain locus, is associated with follicular lymphoma. In most follicular lymphomas, neoplastic germinal centers express high levels of BCL-2 alpha protein, while normal centers are negative. Two transcript variants of BCL-2 differ at their C-terminal ends. Overexpression of BCL-2 is linked to human cancers including B-cell lymphoma and prostate cancer.
