Immunogen

This antibody was prepared by repeated immunizations with recombinant human IL-1 beta produced in E. coli.
The recombinant 153 aa IL-1 beta has a molecular weight of 17 kDa, with the N-terminal amino acid at position alanine 117.
This cleavage site is generated by the IL-1 beta converting enzyme (ICE, caspase-1).

Product Specific Information

• Store vial at 4°C prior to restoration. For extended storage, aliquot and freeze at -20°C or below. Avoid freeze/thaw cycles.
• Centrifuge if solution is not clear after standing. Stable for several weeks at 4°C as undiluted liquid.
• Heated to 56°C for 30 minutes; purified by DEAE fractionation.
• Primarily directed against mature 17 kDa human IL-1 beta; also detects primate IL-1 beta.
• Does not recognize human IL-1 alpha.
• In ELISA assays, detects mainly the mature 17 kDa form and partially the native 31 kDa precursor.
• In immunoblots, detects both 17 kDa (mature) and ~35 kDa (denatured precursor) bands.

Target Information

Interleukin-1 beta (IL-1β) is a proinflammatory cytokine expressed by monocytes, macrophages, and dendritic cells.
It is synthesized as a 31 kDa inactive pro-form and processed into an active 17 kDa protein via inflammasome activation and caspase-1 cleavage.
IL-1β lacks a signal peptide and is secreted through a non-classical pathway.
It signals through IL-1RI and IL-1RII, shared with IL-1α, and its activity can be inhibited by IL-1 Receptor Antagonist (IL-1RA).
IL-1β plays a key role in innate host defense and inflammation; elevated levels are linked to chronic inflammatory diseases.
