Target Information

This gene encodes a transmembrane receptor, commonly referred to as DC-SIGN, expressed on the surface of dendritic cells and macrophages. The protein plays a key role in the innate immune system by recognizing a wide range of pathogens including parasites and viruses.

The protein structure consists of:

  • N-terminal transmembrane domain
  • Tandem-repeat neck domain
  • C-type lectin carbohydrate recognition domain

The extracellular region mediates both pathogen recognition and cell adhesion by binding carbohydrate ligands on microbial and host cell surfaces. The neck domain enables receptor oligomerization, enhancing ligand binding avidity.

Variations in the neck domain repeat number are rare but significantly affect ligand-binding capacity. DC-SIGN is closely related to L-SIGN (GeneID 10332), with differences in ligand-binding and tissue distribution. Alternative splicing produces multiple isoforms.
