TRAF proteins share homology regions enabling receptor binding and activation of signaling cascades.
TRAF2 and TRAF3 bind directly to CD40, essential for NF-κB activation in B lymphocytes.
TRAF2 and TRAF6 are required for CD40 signaling in nonhematopoietic cells.
TRAF2 also interacts with lymphotoxin-beta receptor (LTβR) with TRAF3 and apoptosis inhibitors cIAP1 and Smac.
