Product Specific Information

Description:
The SJ25C1 monoclonal antibody reacts with human CD19, a 95 kDa transmembrane glycoprotein expressed on B cells throughout development (except plasma cells) and on follicular dendritic cells. CD19 forms a complex with CD21, CD81, Leu13, and MHC class II, associating with the BCR to mediate signaling that induces phosphorylation, calcium flux, and proliferation of B cells. SJ25C1 and HIB19 recognize overlapping epitopes.

Each product includes one vial of NovaFluor conjugate and one vial of CellBlox Plus Blocking Buffer.

Applications Reported: Flow cytometric analysis.
Applications Tested: Pre-titrated and validated for flow cytometry using human peripheral blood cells. Recommended use: 5 µL (0.8 µg) per test in 100 µL total volume. Adjust cell number empirically (10⁵–10⁸ cells/test).

Note:
NovaFluor dyes are incompatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI). Use Invitrogen LIVE/DEAD Fixable Dead Cell Stains instead.

CellBlox Plus Blocking Buffer (Cat. No. C001T06F01)

• Required for optimal staining with NovaFluor conjugates
• Add 5 µL per sample to antibody cocktails before combining with cells
• For single-color controls: add 5 µL per 100 µL of cell sample (10³–10⁸ cells)

Target Information

CD19 is a member of the immunoglobulin superfamily with two Ig-like domains. It is expressed on all peripheral B cells and certain myeloid leukemia cells. CD19 functions as a signal-amplifying coreceptor with the B cell receptor (BCR) and plays a key role in regulating B cell activation, proliferation, and differentiation. Mutations can cause hypogammaglobulinemia, while overexpression leads to B cell hyperactivity.
