Product Specific Information

The eBio1D3 (1D3) monoclonal antibody reacts with mouse CD19, a 95 kDa transmembrane glycoprotein expressed by B cells during all developmental stages except terminally differentiated plasma cells. Follicular dendritic cells also express CD19. CD19 forms a multimolecular complex with CD21, CD81, and MHC class II that associates with the BCR, inducing tyrosine phosphorylation, calcium flux, and B cell proliferation.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported:
Suitable for flow cytometric analysis. Internal testing shows NovaFluor Yellow 570 may non-specifically stain B cells in SJL mice but not in BALB/c or C57BL/6 strains.

Applications Tested:
Validated in flow cytometric analysis of mouse splenocytes. Recommended ≤0.2 µg per test (100 µL final volume). Cell number can range from 10⁵ to 10⁸ cells/test. Titrate for optimal performance.

Compatibility Notes:
NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI). Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.

CellBlox Plus Blocking Buffer Usage:

• Required for optimal staining with NovaFluor conjugates.
• Add 5 µL per sample to antibody cocktails before combining with cells.
• For single-color controls, use 5 µL per 100 µL of cell sample containing 10³–10⁸ cells.

NovaFluor Technology:
Based on Phiton™ technology utilizing nucleic acid dye structures engineered for controlled fluorescence signatures, minimizing spillover and spread impacts.

Target Information

CD19 is a member of the immunoglobulin superfamily with two Ig-like domains. It is expressed on 100% of peripheral B cells and on certain myeloid leukemia cells. CD19 acts as a coreceptor for B cell receptor (BCR) signaling, regulating normal and malignant B cell proliferation. Mutation leads to hypogammaglobulinemia, while overexpression causes B cell hyperactivity.
