Product Specific Information

The eBio1A6 monoclonal antibody reacts with human thymic stromal-derived lymphopoietin receptor (TSLPR). TSLPR is an approximately 50 kDa protein similar to the common gamma-chain. It complexes with IL-7R alpha (CD127) to form a high-affinity receptor for TSLP. Human TSLPR is expressed by monocytes and CD11c+ dendritic cells, inducing Th2 cell-attracting chemokines CCL17 and CCL22. Activation of dendritic cells by TSLPR increases secretion of Th cytokines IL-4, IL-5, and IL-13, regulating CD4+ T cell homeostasis.

In mice, TSLPR deficiency alone does not affect lymphocyte numbers, but double deficiency with the common gamma-chain reduces lymphocyte counts compared to gamma-chain deficiency alone. The eBio1A6 monoclonal antibody can cross-block binding of another anti-human TSLPR monoclonal antibody, 1D8.

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and tested on TSLPR-transfected cells. Recommended use: 5 µL (0.25 µg) per test (100 µL final volume). Cell number range: 10⁵–10⁸ cells/test.

Super Bright 600 is a tandem dye excited by a 405 nm violet laser, emitting at 600 nm. Use a 610/20 bandpass filter and ensure instrument compatibility.

When using multiple Super Bright dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific polymer interactions. Refer to the buffer datasheet for details.

Light Sensitivity: Protect vial and samples from light.
Fixation: Samples can be stored in IC Fixation Buffer (Product # 00-8222) or 1-step Fix/Lyse Solution (Product # 00-5333) for up to 3 days at 2–8°C in the dark with minimal impact on brightness and FRET efficiency. Clone-specific performance should be verified empirically.

Target Information

Thymic stromal lymphopoietin (TSLP) drives dendritic cell maturation and activation, supports regulatory T cell selection, peripheral CD4+ T cell homeostasis, and allergic CD4+ T cell responses. It also promotes B cell progenitor growth and differentiation. TSLP receptor (TSLP-R) binds TSLP with low affinity; co-expression with IL-7R-alpha forms a high-affinity, functional heteromeric complex requiring Stat5 for signal transduction, inhibited by SOCS-1.
