Product Specific Information

The 8.1.1 monoclonal antibody reacts with mouse podoplanin (T1a, gp38, aggrus), a 43 kDa transmembrane glycoprotein expressed in kidney podocytes, epithelial, and mesothelial cells. It serves as a potent marker for lymphatic endothelium and is also expressed in thymic subcapsular epithelial cells. Mice deficient in podoplanin exhibit respiratory defects and congenital lymphedema due to lymphatic patterning failure.

Applications Reported: Suitable for flow cytometric analysis.
Applications Tested: Tested on TE-71 cell line; ≤0.125 µg/test recommended. A test defines the antibody amount for staining a 100 µL cell sample (10⁵–10⁸ cells/test).

Fluorochrome Information:

• Super Bright 600 is a tandem dye excited by 405 nm (violet laser) and emits at 600 nm.
• Recommended filter: 610/20 bandpass.
• Use Super Bright Complete Staining Buffer (Product # SB-4401) when using multiple Super Bright dyes to minimize polymer interactions.
• Protect vial and samples from light (photo-oxidation sensitive).
• Samples can be fixed using IC Fixation Buffer (Product # 00-8222) or 1-step Fix/Lyse Solution (Product # 00-5333) and stored up to 3 days at 4°C in the dark.

Target Information

Podoplanin (PDPN, T1A, gp38, Aggrus) is a mucin-type glycoprotein (40–43 kDa) expressed in lymphatic epithelial cells, follicular dendritic cells, and various tumors. It localizes in stromal cells and thymic epithelial cells, contributing to lymphatic endothelium regulation and podocyte morphology. Podoplanin acts as a CLEC-2 ligand and correlates with tumor invasiveness. It is involved in cell migration, metastasis formation, and tumor invasion. Expression is induced via Src-Cas activation and found on Th17 cells. Dysfunction is linked to lymphangioma and subglottis neoplasm, and it is proposed as a marker for lung injury.
