Product Specific Information

The monoclonal antibody 2D3 reacts to CD47, also known as integrin-associated protein (IAP) and neurophilin.
CD47 is a glycosylated five-transmembrane protein involved in adhesion through interactions with SIRP (signal regulator protein) and associated with beta3 integrins (CD51/CD61 and CD41/CD61).
It mediates bi-directional signaling to regulate neural synaptic activity and macrophage phagocytosis. CD47 also serves as a receptor for thrombospondin and is expressed in most hematopoietic cells (T, B, monocytes, platelets, erythrocytes) and non-hematopoietic cells.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Suitable for flow cytometric analysis.
Applications Tested: Pre-titrated and tested on normal human peripheral blood cells.
Use at 5 µL (0.6 µg) per test. A test is defined as the amount of antibody that stains a sample in 100 µL final volume (10⁵–10⁸ cells/test).

Note:
NovaFluor dyes are not compatible with DNA intercalating viability dyes such as propidium iodide, 7-AAD, and DAPI.
Use Invitrogen LIVE/DEAD Fixable Dead Cell stains with NovaFluor dyes.

CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining.
Add 5 µL per sample to antibody cocktails before combining with cells.

NovaFluor conjugates utilize Phiton™ technology, enabling engineered fluorescent signatures with minimized spillover and spread impacts.

Target Information

CD47 (integrin-associated protein, IAP) is a transmembrane glycoprotein interacting with integrins and regulating their functions.
It modulates signaling pathways, including activation of heterotrimeric G proteins, and increases intracellular calcium upon cell adhesion.
CD47 is a receptor for thrombospondin and may play roles in membrane transport and signal transduction.
It is broadly expressed, reduced on Rh erythrocytes, and acts as a ligand for CD172a (SIRPα), preventing phagocytosis of CD47-positive cells.
CD47-CD172a interaction influences cell migration, B cell adhesion, T cell activation, chondrocyte response, and neuronal development.
