Description:
The eBio156-4H9 monoclonal antibody reacts with the human CD48 antigen, also known as B cell activation marker 1, SLAM family member 2, Blast-1 (human), and OX-45 (rat). CD48, part of the SLAM family and Ig superfamily, is a 45 kDa GPI-linked glycoprotein expressed on most hematopoietic cells. CD2 is the low affinity ligand for CD48.

Recent studies show differential expression of SLAM family members (CD48, CD150, CD244) among distinct bone marrow hematopoietic progenitors, providing a useful tool for predicting progenitor primitiveness. CD48 is expressed on many leukocytes, minimally on granulocytes, and absent on platelets and erythrocytes. Expression increases upon B cell activation.

Reported for use in flow cytometric analysis and functional assay.

Applications Tested:
Tested by flow cytometric analysis of normal human peripheral blood cells. Recommended ≤0.5 µg per test (100 µL final volume). Cell number may vary (10⁵–10⁸ cells/test). Optimize antibody titration for best performance.

Target Information

CD48 (Blast-1) belongs to the CD2 subset of the Ig superfamily, including CD2, CD58, CD84, CD150, CD229, CD244, etc. These molecules mediate homotypic or heterotypic adhesion through interactions within the Ig family.

CD48 is a GPI-anchored protein broadly expressed on hematopoietic cells and serves as a high affinity ligand for 2B4 and low affinity ligand for CD2. The 2B4-CD48 interaction among NK and NK-T cells regulates proliferation. CD48 signaling activates eosinophils and is upregulated in infectious diseases.

Strongly expressed on lymphocytes and monocytes, weakly on granulocytes, and absent on platelets, fibroblasts, epithelium, and endothelium. CD48 is a marker for detecting GPI anchoring defects in paroxysmal nocturnal hemoglobinuria (PNH) and may serve as a pharmacological target in allergic airway diseases such as rhinitis and asthma.
