Target Information
Upon entry into mitosis, the cell’s microtubule (MT) network forms the mitotic spindle, enabling segregation of paired chromosomes. Kinetochores (KT) on centromeric chromatin are essential for proper attachment of chromosomes to spindle MTs.
The spindle and kinetochore complex (SKA complex), composed of SKA1, SKA2, and SKA3, is necessary for stable KT–MT interactions and timely anaphase onset.
Depletion of SKA1 or SKA2 by siRNA removes both proteins from KT without affecting overall KT structure. SKA complex depletion causes a prolonged checkpoint-dependent metaphase delay, highlighting its role in metaphase plate maintenance and spindle checkpoint silencing.
SKA2 also interacts with glucocorticoid receptors, contributing to glucocorticoid signaling and cell proliferation.
