Target Information

RIPK1 is a serine-threonine kinase that transduces inflammatory and cell-death signals following death receptor ligation, activation of pathogen recognition receptors (PRRs), and DNA damage.
Upon activation of TNFR1 by TNF-alpha family cytokines, TRADD and TRAF2 are recruited to the receptor. It phosphorylates DAB2IP at Ser-728 in a TNF-alpha-dependent manner, activating the MAP3K5-JNK apoptotic cascade.
Ubiquitination by TRAF2 via Lys-63-linked chains enhances downstream signaling through MAPK and NF-kappa-B pathways, leading to expression of inflammatory molecules.
Polyubiquitinated RIPK1 binds to IKBKG/NEMO, a key step for NF-kappa-B activation. Interaction with RHIM-containing adapters triggers gene activation and cell death. RIPK1 and RIPK3 association forms a necrosis-inducing complex.
