Product Specific Information

PEP-016 is a 15 amino acid (amino acids 533–547) synthetic peptide whose sequence is found N-terminal to the PR DNA binding domain.
Sequence (amino to carboxy terminus): G-L-P-Q-V-Y-P-P-Y-L-N-Y-L-R-P

This peptide may be used for neutralization and control experiments with the monoclonal antibody that reacts with this product and PR (catalog # MA1-410). Using a solution with equal weights per unit volume of peptide and corresponding antibody will yield a solution with a large molar excess of peptide capable of competitively binding the antibody.

Reconstitution: Add 0.1 mL of distilled water.

Target Information

The progesterone receptor (PR) is a member of the steroid family of nuclear receptors. It mediates the physiological effects of progesterone, which plays a central role in reproductive events associated with pregnancy establishment and maintenance.

PR exists as:

  • Form A (94 kDa)
  • Form B (120 kDa)

These forms arise from alternative translation initiation sites. PR-B is transcriptionally active and responsible for activating genes for endometrial maintenance, pregnancy maintenance, and ovulation inhibition.
PR-A is identical to PR-B except for a 165 amino acid deletion at the N-terminus, exposing a 140 amino acid inhibitory domain (ID) that represses steroid hormone transcriptional activity.

In its inactive state, PgR forms a multiprotein complex including heat shock proteins and immunophilins. Upon progesterone binding, conformational change allows receptor dimerization and binding to progesterone response elements (PRE), resulting in activated transcription.
A null mutation in the PGR gene leads to pleiotropic reproductive abnormalities.
