Target Information
Protein tyrosine phosphatases (PTPs) are enzymes that remove phosphate groups from phosphorylated tyrosine residues on proteins. Together with tyrosine kinases, they regulate the phosphorylation state of many signaling molecules, such as the MAP kinase family.
PTPs are involved in cell signaling, growth, proliferation, and oncogenic transformation. Dual specificity phosphatases (DSPs), including DUSP3, are a subclass of PTPs that dephosphorylate both phosphothreonine and phosphotyrosine residues. The VH1 gene in vaccinia virus is the prototypical DSP, with a shallow active site pocket allowing hydrolysis of phosphorylated serine, threonine, or tyrosine residues.
