Target Information
The mediator complex is a multi-protein transcriptional co-activator expressed ubiquitously in eukaryotes and required for RNA polymerase II transcription activation. MED28 (also known as Magicin) is one of the proteins in this complex and is expressed in various cell types and tissues.
Down-regulation of MED28 in NIH3T3 cells induces smooth muscle cell differentiation genes, while over-expression represses them. MED28 can form a ternary complex with Grb2 and Merlin (NF2 tumor suppressor), suggesting a role in tumor suppression.
Additionally, MED28 has been identified as an HIV dependency factor (HDF), indicating potential as a drug target in HIV treatment. At least two isoforms of MED28 exist.
