Product Specific Information

The OKT8 monoclonal antibody reacts with human CD8a, a 32–34 kDa cell surface receptor expressed as a heterodimer (CD8αβ) or homodimer (CD8αα).
CD8a is found on thymocytes, mature T cells, and NK cells, and binds MHC class I molecules. This interaction, via protein tyrosine kinase p56lck, contributes to T-cell development and activation.

OKT8 does not compete with other mouse anti-human CD8 clones (RPA-T8, HIT8a) for binding to human leukocytes, indicating distinct epitopes.

Each product includes:

  • 1 vial of NovaFluor conjugate
  • 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and validated for human peripheral blood cells at 5 µL (0.1 µg)/test.
A test is defined as staining in a final volume of 100 µL with 10⁵–10⁸ cells/test.

NovaFluor Compatibility Notes:

  • Not compatible with DNA intercalating viability dyes (e.g., PI, 7-AAD, DAPI).
  • Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability.

CellBlox Plus Blocking Buffer (C001T06F01):

  • Improved formulation over CellBlox.
  • Required for optimal NovaFluor staining.
  • Add 5 µL per sample to antibody cocktails or single-color controls.

Target Information

CD8 is a type I transmembrane glycoprotein in the immunoglobulin receptor family, crucial for signal transduction and T-cell differentiation.
It acts as a co-receptor with TCR for MHC class I molecules, while CD4 acts for MHC class II.
CD8α binds MHC-I and is expressed on T cells, NK cells, monocytes, and dendritic cells.
Engagement of MHC-I/peptide complexes recruits Lck, leading to cytokine production and cytotoxic T lymphocyte (CTL) activation.
Activated CTLs are essential for pathogen and tumor cell clearance. Differentiation of naive CD8+ T cells into CTLs is enhanced by IL-2, IL-12, and TGF-β1.
