Product Specific Information

The OKT8 monoclonal antibody reacts with the human CD8a molecule (32–34 kDa), expressed as a heterodimer (CD8αβ) or homodimer (CD8αα). CD8a is found on most thymocytes, a subset of mature T cells, and NK cells. It binds to MHC class I and associates with protein tyrosine kinase p56lck, contributing to T-cell development and activation.

Preliminary testing indicates OKT8 does not compete with other anti-human CD8 antibodies (RPA-T8, HIT8a) for binding, suggesting distinct epitopes.

Each product includes:

  • 1 vial of NovaFluor conjugate
  • 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and validated for flow cytometry of normal human peripheral blood cells. Use at 5 µL (0.2 µg)/test in a final volume of 100 µL. Recommended cell count: 10⁵–10⁸ cells/test.

Important Notes:

  • NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., PI, 7-AAD, DAPI).
  • Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for compatibility.
  • CellBlox Plus Blocking Buffer is required for optimal staining. Add 5 µL per sample to antibody cocktails before combining with cells.
  • For single-color controls: use 5 µL blocking buffer per 100 µL of cell sample (10³–10⁸ cells).

NovaFluor conjugates utilize Phiton™ technology, employing nucleic acid dye structures for engineered fluorescent signatures that minimize spillover and spread.

Target Information

CD8 (Cluster of Differentiation 8) is a type I transmembrane glycoprotein in the immunoglobulin receptor family. It plays a crucial role in T-cell signal transduction, differentiation, and activation.

  • Expressed on T cells as a CD8αβ heterodimer or CD8αα homodimer on subsets of T cells, NK cells, monocytes, and dendritic cells.
  • Functions as a co-receptor with the TCR for MHC class I molecules.
  • Engagement of MHC-I/peptide complexes recruits Lck kinase, leading to CTL activation and pathogen/tumor clearance.
  • Differentiation of naive CD8⁺ T cells into cytotoxic T lymphocytes (CTLs) is enhanced by IL-2, IL-12, and TGF-β1.
