Product Specific Information
The OKT8 monoclonal antibody reacts with human CD8a, a 32–34 kDa cell surface receptor expressed as CD8αβ heterodimers or CD8αα homodimers. CD8a is found on most thymocytes and subsets of mature T and NK cells. CD8 binds to MHC class I and, via p56lck kinase association, contributes to T-cell development and activation.
Each product includes one vial of NovaFluor conjugate and one vial of CellBlox Plus Blocking Buffer.
This antibody is optimized for flow cytometric analysis and pre-titrated for use at 5 µL (0.1 µg)/test in a 100 µL final volume. Cell count per test may range from 10⁵ to 10⁸.
Note:
- NovaFluor dyes are not compatible with DNA intercalating dyes (e.g., propidium iodide, 7-AAD, DAPI).
- Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability.
- CellBlox Plus Blocking Buffer (Product # C001T06F01) is required for optimal staining. Add 5 µL per sample before combining with cells.
NovaFluor conjugates utilize Phiton™ technology, providing engineered fluorescent signatures designed to minimize spillover and spread.

Target Information
CD8 (Cluster of Differentiation 8) is a type I transmembrane glycoprotein of the immunoglobulin receptor family involved in signal transduction, T-cell differentiation, and activation.
- Expressed primarily on T cells as CD8αβ heterodimers, functioning as a co-receptor for MHC class I molecules.
- CD8α chain binds directly to MHC-I, while CD8αα homodimers are found on subsets of T cells, NK cells, monocytes, and dendritic cells.
- Ligation with MHC-I/peptide complexes triggers Lck kinase recruitment, lymphokine production, and cytotoxic T lymphocyte (CTL) activation.
- Differentiation of naive CD8⁺ T cells into CTLs is enhanced by IL-2, IL-12, and TGF-β1.
