The monoclonal antibody 5B10 recognizes mouse Caspase-1, a cysteine-dependent aspartate protease crucial in apoptosis. Caspase-1 converts pro-IL-1β to its mature form, also known as ICE (interleukin-1β converting enzyme). Upon activation, Caspase-1 forms a tetramer of two p20 and two p10 subunits, with multiple isoforms identified.

The 5B10 antibody detects both the pro-form (~42 kDa) and the active p20 form of Caspase-1.

Target Information

Caspases are a family of cysteine proteases divided into apoptotic and inflammatory subfamilies. Unlike apoptotic caspases, inflammatory caspases (including Caspase-1, -4, -5, and -12) are primarily involved in immune responses to microbial pathogens. They activate proinflammatory cytokines such as IL-1β and IL-18.

Caspase-1 was first identified as an IL-1β-converting enzyme, later found to be a mammalian homolog of the C. elegans cell death gene ced-3, whose overexpression induces apoptosis in fibroblasts.
