Product Specific Information

The 145-2C11 monoclonal antibody reacts with mouse CD3e, a 20 kDa subunit of the TCR complex. Along with CD3 gamma and delta, the epsilon chain is required for proper assembly, trafficking, and surface expression of the TCR complex. CD3 is expressed by thymocytes in a developmentally regulated manner and by all mature T cells.

Binding of 145-2C11 to TCR initiates intracellular signaling leading to activation, proliferation, or apoptosis depending on conditions. Commonly used as a phenotypic marker for mouse T cells.

Brilliant Violet™ 711 (BV711) is a tandem dye emitting at 713 nm, suitable for cytometers equipped with a violet (405 nm) laser. Ensure instrument compatibility.

Buffer Recommendation:
When using multiple Brilliant Violet™, Brilliant Ultra Violet™, or polymer dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401-42) or Brilliant Stain Buffer™ (Product # 00-4409-75) to minimize non-specific interactions.

Light Sensitivity: Protect vial and samples from light.
Fixation: Compatible with IC Fixation Buffer (Product # 00-8222-49) or 1-step Fix/Lyse Solution (Product # 00-5333-54) for up to 3 days at 4°C in the dark. Not compatible with methanol-based fixation.

Target Information

The CD3 complex transduces antigen-recognition signals into T cells and regulates TCR surface expression. It includes CD3 gamma, delta, epsilon, and zeta subunits, encoded by genes on chromosome 11. These subunits associate with signal transduction molecules via tyrosine-based motifs.

CD3 is expressed on 68–82% of normal peripheral blood lymphocytes, 65–85% of thymocytes, and Purkinje cells, but not on B or NK cells. Defects in CD3 genes are linked to CD3 immunodeficiency.
