Product Specific Information

Description:
The O323 monoclonal antibody reacts with human CD27, a lymphocyte-specific member of the TNFR superfamily. CD27 is expressed by a subset of thymocytes and virtually all mature T cells and is upregulated upon T-cell stimulation. CD27 binds to CD70 and plays an important role in T cell–B cell interaction.

Applications Reported:
This O323 antibody is reported for use in flow cytometric analysis.

Applications Tested:
Pre-titrated and tested by flow cytometric analysis of normal human peripheral blood cells. Recommended use is 5 µL (0.25 µg) per test (100 µL final volume). Cell number may vary from 10⁵ to 10⁸ cells/test.

Fluorochrome Information:
Super Bright 436 is excited with the violet laser (405 nm) and emits at 436 nm. Recommended filter: 450/50 bandpass or equivalent. Ensure instrument compatibility.

When using multiple Super Bright dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific polymer interactions.

Target Information

CD27 is a 50 kDa member of the TNF receptor superfamily (includes CD40, CD30). It regulates cell proliferation and death. Expression of CD27 and its ligand CD70 is confined to lymphocytes. CD27 is a disulfide-linked homodimer on thymocytes, peripheral T cells, and some B cells. Upon T-cell activation, CD27 expression increases and soluble CD27 (28–32 kDa) is released into plasma, urine, or spinal fluid, serving as a prognostic marker in B-cell malignancies.

RgpA, a cysteine proteinase, can degrade CD27 and counteract T-cell activation. CD27-binding protein (SIVA) is a proapoptotic protein involved in CD27-mediated apoptosis.
Diseases associated with CD27 dysfunction include Lymphoproliferative Syndrome 2 and Autosomal Recessive Lymphoproliferative Syndrome.
