Preparation:
Produced from sera of rabbits immunized with highly pure Recombinant Human Leptin. Anti-Human Leptin-specific antibody was purified by affinity chromatography employing an immobilized Human Leptin matrix.

Sandwich ELISA:
To detect Human Leptin by sandwich ELISA (using 100 µL/well antibody solution), a concentration of 0.5–2.0 µg/mL is required. This antigen affinity purified antibody, in conjunction with PeproTech Biotinylated Anti-Human Leptin (500-P86Bt) as a detection antibody, allows detection of at least 0.2–0.4 ng/well of Recombinant Human Leptin.

Western Blot:
To detect Human Leptin by Western Blot analysis, this antibody can be used at a concentration of 0.1–0.2 µg/mL. Used with compatible secondary reagents, the detection limit for Recombinant Human Leptin is 1.5–3.0 ng/lane under either reducing or non-reducing conditions.

Target Information

Leptin is a 167 amino acid protein hormone with key roles in regulating body weight, metabolism, and reproductive function. It has a molecular weight of approximately 16 kDa and is encoded by the obese (ob) gene. Leptin is mainly secreted by adipocytes, with smaller amounts produced by stomach epithelium and placenta.
Leptin receptors are highly expressed in hypothalamic regions that regulate feeding behavior, hunger, body temperature, and energy expenditure. It is involved in hematopoiesis, angiogenesis, wound healing, inflammation, immune responses, pubertal development, and fetal growth.
Studies have linked leptin to obesity, anorexia nervosa, insulin resistance, and hypertension. It also regulates fatty acid oxidation enzymes and thermogenesis. Defects in leptin production can cause severe hereditary obesity.
