Product Specific Information
This antibody was cross-adsorbed to phospho-WAVE (Tyr-125) and unphosphorylated WAVE (Tyr-150) peptides before affinity purification using phospho-WAVE (Tyr-150) peptide.
In western blots, the antibody detects a 78 kDa band corresponding to phosphorylated WAVE2 in K562 cells treated with pervanadate and an 80 kDa band corresponding to WAVE1 in HUVEC treated with pervanadate. It also recognizes an unidentified band near 50 kDa in K562 cells.
Target Information
WASP (Wiskott-Aldrich syndrome protein) and N-WASP are downstream effectors of Cdc42 that are implicated in actin polymerization and cytoskeletal organization.
The WASP family also includes VASP and Mena, which accumulate at focal adhesions and regulate actin cytoskeleton.
WAVE proteins are related to WASP family and mediate actin reorganization downstream of Rho family GTPases.
WAVE1 and WAVE2 specifically regulate membrane ruffling by inducing actin filament clusters in response to GTP binding and Rac activation.
These proteins cooperate with the Arp2/3 complex to promote actin filament formation.
WAVE1 (also SCAR) is expressed primarily in the brain; WAVE2 is widely expressed, highest in peripheral blood leukocytes.
WAVE3 forms a multiprotein complex linking receptor kinases and actin via a C-terminal verprolin homology domain.
This complex transduces signals involving changes in cell shape, motility, or function.
