Product Specific Information
- Specificity: R-125224 binds to the extracellular portion of human Fas at an epitope consisting of the sequence RTQNTKCRCK (aa 105–114).
- Preparation: R-125224 is generated by humanizing the murine HFE7A anti-Fas antibody through grafting the CDR regions to the framework regions of the human 8E10 antibody and substituting key framework residues from the murine antibody into the 8E10 sequence.
The original HFE7A was derived from a hybridoma cell line generated by fusing NS1 myeloma cells with splenocytes from Fas-deficient mice immunized with partially purified recombinant human Fas-AIC2A chimera protein. The HFE7A hybridoma was selected via flow cytometry for antibodies binding to WR19L12a (murine T cell lymphoma expressing human Fas) or L5178YA1 (expressing murine Fas), but not to parental WR19L or L5178Y cells.
Target Information
FAS (Fas, CD95, APO-1) is a 46 kDa transmembrane glycoprotein functioning as a cell death receptor of the TNFR superfamily. A soluble form (~26 kDa) also exists.
Stimulation of FAS triggers aggregation of FAS-associated death domains (FADD), formation of the death-inducing signaling complex (DISC), and activation of caspases, leading to apoptosis.
FAS can activate NF-kappaB, MAPK3/ERK1, and MAPK8/JNK pathways, participating in proliferative signaling in fibroblasts and T cells.
Multiple alternatively spliced transcript variants exist, with isoforms lacking the transmembrane domain acting as negative regulators of apoptosis.
The Fas/Fas ligand system is implicated in diseases such as AIDS, hepatitis, and cancer, and plays a role in antiviral immune responses.
